Single Step, Real-Time Protein Kinase Detection For Biological Screening

ABSTRACT:

Scientists at Berkeley Lab and the University of California have developed a kinase profiling assay and detection system that can acquire kinetic information of real-time phosphorylation reactions using a nanoengineered microarray of kinase substrates.

Conventional kinase profiling and kinetic screening can only detect phosphorylation levels at reaction endpoints.  The disadvantages are that these conventional methods rely on either radioactive kinase assays or detecting fluorescent molecules. Also the ability to screen a wide array of kinase substrates is often time consuming and expensive.

In Berkelely Lab’s screening method, Fanqing Frank Chen and colleagues have conjugated kinase-specific substrates to a nanoengineered microarray and measured kinase activity by monitoring phosphorylation levels.  Biological samples are introduced to the detection system and simultaneous multiplexed kinase screening can take place as the signal is monitored to detect phosphorylation levels in real-time.
